We have developed innovative assays that can detect enzymes rapidly. Paracetamol- or catechol-bearing compounds, when exposed to their respective enzymes, released paracetamol or catechol, which can be detected using a standard glucose meter. This approach was used to detect a number of diverse analytes that include enzymes such as beta-galactosidase and alpha-mannosidase and pathogens such as influenza viruses, Streptococcus pneumoniae, and E. coli rapidly. The limit of detection for all analytes was extremely low and clinically relevant for influenza viruses. We also demonstrate that glucose oxidase or glucose dehydrogenase is not required because the paracetamol gets oxidized directly on the electrode surface. This indicates that test strips without glucose oxidase or dehydrogenase can be used, and we can detect analytes in the presence of high levels of background glucose. We demonstrate this unique nature of the assay to detect paracetamol in simulated urine and sheep blood without background interference of intrinsic glucose, indicating that glucose meters can be used to detect nonglucose analytes without background glucose interference.

A simple procedure leading to disposable potentiometric sensors using as a supporting electrode – electrical lead and transducer – a layer of carbon nanostructured material, either graphene or multiwalled nanotubes, is proposed, and the effect of the material used on the properties of the sensor is discussed. The obtained layers were partially covered with a conventional poly(vinyl chloride) (PVC) based ion-selective membrane to result in simple, planar, and disposable potentiometric sensors. The analytical performance of the thus obtained electrodes was compared with that of classical macroscopic all-solid-state ion-selective electrodes (e.g. employing poly(octylthiophene) as a solid contact and a similar ion-selective membrane). It was superior (taking into account detection limits or selectivity towards Na+ ions) compared to that of other disposable sensors proposed recently. The observed excellent analytical performance was attributed to the applied method of preparation of carbon nanostructured materials, which does not require addition of a surfactant to obtain a stable suspension (ink) used to prepare the electrical lead and the transducer of the sensor. Although the proposed sensors are predominantly intended for disposable use, pronounced stability of potential readings was obtained in within-day experiments. Moreover, due to their high conductivity carbon-plastic electrodes can be also applied in polarized potentiometric measurements.

US28 is a human cytomegalovirus (HCMV) encoded G-protein-coupled receptor that signals in a constitutively active manner. Recently, we identified 1 {5-(4-(4-chlorophenyl)-4-hydroxy-piperidin-1-yl)-2,2-diphenylpentanenitrile} as the first reported nonpeptidergic inverse agonist for a viral-encoded chemokine receptor. Interestingly, this compound is able to partially inhibit the viral entry of HIV-1. In this study we describe the synthesis of 1 and several of its analogues and unique structure-activity relationships for this first class of small-molecule ligands for the chemokine receptor US28. Moreover, the compounds have been pharmacologically characterized as inverse agonists on US28. By modification of lead structure 1, it is shown that a 4-phenylpiperidine moiety is essential for affinity and activity. Other structural features of 1 are shown to be of less importance. These compounds define the first SAR of ligands on a viral GPCR (US28) and may therefore serve as important tools to investigate the significance of US28-mediated constitutive activity during viral infection.

Based upon a previously reported lead compound 1, a series of 1,2-diamino-ethane-substituted-6,7,8,9-tetrahydro-5H-pyrimido[4,5-d]azepines were synthesized and evaluated for improved physiochemical and pharmacokinetic properties while maintaining TRPV1 antagonist activity. Structure-activity relationship studies directed toward improving the aqueous solubility (pH 2 and fasted-state simulated intestinal fluid (SIF)) and rat pharmacokinetics led to the discovery of compound 13. Aqueous solubility of compound 13 (pH 2 = >237 mug/mL and SIF = 11 mug/mL) was significantly improved over compound 1 (pH 2 = 5 mug/mL and SIF = 0.5 mug/mL). In addition, compound 13 afforded improved rat pharmacokinetics (CL = 0.7 L/kg/h) compared to compound 1 (CL = 3.1 L/kg/h). Compound 13 was orally bioavailable and afforded a significant reversal of carrageenan-induced thermal hyperalgesia at 5 and 30 mg/kg in rats.

To develop novel CDK2 inhibitors as anticancer agents, a series of novel pyrimidine-based benzothiazole derivatives were designed and synthesized. Initial biological evaluation demonstrated some of target compounds displayed potent antitumor activity in vitro against five cancer cell lines. Especially, the analogue 10s exhibited approximately potency with AZD5438 toward four cells including HeLa, HCT116, PC-3, and MDA-MB-231 with IC50 values of 0.45, 0.70, 0.92, 1.80 muM, respectively. More interestingly, the most highly active compound 10s in this study also possessed promising CDK2/cyclin A2 inhibitory activities with IC50 values of 15.4 nM, which was almost 3-fold potent than positive control AZD5438, and molecular docking studies revealed that the analogue bound efficiently with the CDK2 binding site. Further studies indicated that compound 10s could induce cell cycle arrest and apoptosis in a concentration-dependent manner. These observations suggest that pyrimidine-benzothiazole hybrids represent a new class of CDK2 inhibitors and well worth further investigation aiming to generate potential anticancer agents.
