A Novel Highly Sensitive Method for Measuring Inflammatory Neural-Derived APC Activity in Glial Cell Lines, Mouse Brain and Human CSF

Background: Neural inflammation is linked to coagulation. Low levels of thrombin have a neuroprotective effect, mediated by activated protein C (APC). We describe a sensitive novel method for the measurement of APC activity at the low concentrations found in neural tissue. Methods: APC activity was measured using a fluorogenic substrate, Pyr-Pro-Arg-AMC, cleaved preferentially by APC. Selectivity was assessed using specific inhibitors and activators. APC levels were measured in human plasma, in glia cell lines, in mice brain slices following mild traumatic brain injury (mTBI) and systemic lipopolysaccharide (LPS) injection, and in cerebrospinal fluid (CSF) taken from viral meningoencephalitis patients and controls. Results: Selectivity required apixaban and alpha-naphthylsulphonylglycyl-4-amidinophenylalanine piperidine (NAPAP). APC levels were easily measurable in plasma and were significantly increased by Protac and CaCl2. APC activity was significantly higher in the microglial compared to astrocytic cell line and specifically lowered by LPS. Brain APC levels were higher in posterior regions and increased by mTBI and LPS. Highly elevated APC activity was measured in viral meningoencephalitis patients CSF. Conclusions: This method is selective and sensitive for the measurement of APC activity that significantly changes during inflammation in cell lines, animal models and human CSF.

Molecular, supramolecular, DNA-binding and biological studies of piperazine and piperidine based dithiocarbamates of biocompatible copper

The reaction of dithiocarbamate ligands with CuCl2 center dot 2H(2)O gave the metal complexes of general formula Cu (S2CR)(2), where R = N(CH2)(4)N(Bn) (1), N(CH2)(4)N(MeO-Ph) (2), N(CH2)(4)N(C2H4OH) (3), N(CH2)(4)CHCH(Ph)(2) (4) and N(CH2)(4)CHC(OH)(Ph)(2) (5). The characterization was carried out by elemental analysis and spectro-scopic methods {FT-IR, UV-Visible, NMR (H-1 & C-13) and single crystal XRD}. The XRD data of three square planar complexes (1-3) revealed that 1 and 3 crystalize in monoclinic space group P2(1)/c and C2/c respectively, while 2 in the triclinic group P-1. A variety of interesting supramolecular motifs have been observed for these complexes ranging from 0 and 1-D chains to 2-D layers and 3-D networks. Moreover, DNA binding capability (determined by UV-Visible spectroscopy, cyclic voltammetry and viscometry) and biological activites (anti-oxidant, cytotoxicity and antileishmanial) of the synthesized metal complexes were also investigated. The complexes interacted with DNA base-pairs either through the intercalative or by electrostatic mode (complex 2 in CV experiment) with high binding constant values. As for biological activities are concerned, the synthesized compounds were found promising in all the assays.

Eremomycin pyrrolidide: a novel semisynthetic glycopeptide with improved chemotherapeutic properties

Purpose: Development of new semisynthetic glycopeptides with improved antibacterial efficacy and reduced pseudoallergic reactions. Methods: Semisynthetic glycopeptides 3-6 were synthesized from vancomycin (1) or eremomycin (2) by the condensation with pyrrolidine or piperidine. The minimum inhibitory concentration (MIC) for the new derivatives was measured by the broth micro-dilution method on a panel of clinical isolates of Staphylococcus and Enterococcus. Acute toxicity (50% lethal dose, maximum tolerated doses), antibacterial efficacy on model of systemic bacterial infection with S. aureus and pseudoallergic inflammatory reaction (on concanavalin A) of eremomycin pyrrolidide (5) were evaluated in mice according to standard procedures. Results: The eremomycin pyrrolidide (5) was the most active compound and showed a high activity against Gram-positive bacteria: vancomycin-susceptible staphylococci and enterococci (minimum inhibitory concentrations [MICs] 0.13-0.25 mg/L), as well as vancomycin-intermediate resistant Staphylococcus aureus (MICs 1 mg/L). Antimicrobial susceptibility tested on a panel of 676 isolates showed that 5 had similar activity for the genera Staphylococcus and Enterococcus with MIC90= 0.5 mg/L, while vancomycin had MIC90= 1-2 mg/L. The number of resistant strains of Enterococcus faecium (vancomycin-resistant enterococci) (MIC = 64 mg/L) with this value was 7 (8%) for vancomycin (1) and 0 for the compound 5. In vivo comparative studies in a mouse model of systemic bacterial infection with S. aureus demonstrated that the efficacy of 5 was notably higher than that of the original antibiotics 1 and 2. In contrast to 1, compound 5 did not induce pseudoallergic inflammatory reaction (on concanavalin A). Conclusion: The new semisynthetic derivative eremomycin pyrrolidide (5) has high activity against staphylococci and enterococci including vancomycin-resistant strains. Compound 5 has a higher efficacy in a model of staphylococcal sepsis than vancomycin (1) or eremomycin (2). In striking contrast to natural antibiotics, the novel derivative 5 does not induce a pseudoallergic inflammatory reaction to concanavalin A and therefore has no histamine release activity. These results indicate the advantages of a new semisynthetic glycopeptide antibiotic eremomycin pyrrolidide (5) which may be a prospective antimicrobial agent for further pre-clinical and clinical evaluations.
