Antihistamine drugs are extensively used to treat allergic rhinitis and other allergies. They provide relief from nasal congestion, sneezing, or hives caused by pollen and dust mites with few side effects. Various drugs with different brands and variations in prices are available in the Indian market. This study was done to fiend out the percentage variation of cost among different brands of antihistamine drugs available in the Indian market. The cost of various second-generation antihistamine drugs available in the Indian market were referred from CIMS (current index of medical specialties) (March 2022) and Indian Drug Today (Oct. 2021 to Jan. 2022). The highest and lowest price for ten (10) tablets/capsules of each second-generation antihistamine was analyzed. The percentage cost variation and cost ratio were calculated for each drug. A total of eleven oral Second Generation Antihistamines available in the Indian market and 566 oral tablets or capsules manufactured by different companies were identified for all the Second Generation Antihistamines. Huge variation was found in the cost of different branded preparations for the same drug (40.14% for cetirizine; INR 2.06 to INR 84.76). The most expensive cetirizine was 11.39 times costlier than the least expensive cetirizine. The least-cost variation and cost ratio saw with Bepotastine (10%) and (1.1) resp. In this study, we aimed to investigate the influence of mental health problems developed during the COVID-19 pandemic on the course of mastocytosis. Mental health status in 60 adult patients with mastocytosis was prospectively evaluated with the total Depression-Anxiety-Stress Scale (tDASS-21) and Fear of COVID-19 Scale (FCV-19S) in the lockdown period (LP) and the return to normal period (RTNP) during the pandemic. The disease course was assessed from emergency and outpatient medical reports, including Scoring Mastocytosis (SCORMA) index and serum baseline tryptase levels, by telephone interviews and clin. visits. The mean FCV-19S and median tDASS-21 scores were significantly higher in LP than RTNP (p < 0.001) and there was a pos. correlation between FCV-19S and tDASS-21 in LP (r = 0.820, p < 0.001) and in RTNP (r = 0.572 p= <0.001). Depression is one of the main diseases that lead to disability and loss of ability to work. As a traditional Chinese medicine, Zhi-zi-chi decoction is utilized to regulate and improve depression. In this study, the animal model of depression was established by chronic unpredictable mild stimulation combined with feeding alone. The brain metabolic profile of depressed rats was analyzed by the method of metabolomics based on ultra-performance liquid chromatog.-quadrupole/time-of-flight mass. 26 differential metabolites and six metabolic pathways related to the antidepressant of Zhi-zi-chi decoction were screened and analyzed. The targeted metabolism of the glutathione metabolic pathway was analyzed. The objective of this study was to describe ondansetron drug utilization patterns during pregnancy to treat nausea and vomiting in pregnancy (NVP). Moreover, we aimed to describe the maternal factors associated with NVP and antiemetic use. The data consist of pregnancies with a live birth(s) within an IMRD-UK registered GP practice. Descriptive statistics were used to investigate patterns of ondansetron use in pregnancy and to describe maternal characteristics associated with NVP and antiemetic drug utilization. We differentiate first- from second-line use during pregnancy using antiemetic prescription pathways. The dataset included 733 633 recorded complete pregnancies from 2005 to 2019. NVP diagnosis and ondansetron prescription prevalence increased from 2.7% and 0.1% in 2005 to 4.8% and 2.5% in 2019 resp. Over the period 2015-2019, the most common oral daily dosages were 4 mg/d (8.5%), 8 mg/d (37.1%), 12 mg/d (37.5%) and between 16 and 24 mg/d (16.9%). Prescription of ondansetron was initiated during the first trimester of pregnancy in 40% of the cases and was moderately used as a first-line therapy (2.8%), but preferred choice of second-line therapy. Women with mental health disorders, asthma and/or prescribed folic acid were more likely to experience NVP and use antiemetics in pregnancy than their counterparts. This study confirms that ondansetron is increasingly used off-label to treat NVP during pregnancy, also in the first trimester and before other prescription antiemetics have been prescribed. This study investigates the prediction of removal efficiency of pharmaceuticals (PhCs), personal care products (PCPs), and steroidal hormones (SHs) based on design and operational parameters (depth, area, hydraulic loading rate-HLR, organic loading rate-OLR, and hydraulic retention time-HRT) of constructed wetlands (CWs). A comprehensive statistical anal. was performed by applying principle component anal., correlation and multiple linear regression analyses. The data used in this anal. was compiled from peer reviewed publications. The CWs design and operational parameters are good predictors of the removal efficiency of these emerging organic contaminants. Operational parameters (HLR, OLR, and HRT) are the most significant predicators and combination with design parameters (depth and area) often improved reliability of the predictions. The best predictive models for PhCs and PCPs were composed of depth, OLR, and HRT (root mean square errors-RMSEs: training set: 7-14%; test set: 22-27%). A combination of area, HLR, and OLR formed a credible model for predicting the removal efficiency of SHs (RMSEs: training set: 6%; test set: 11%). Similarly, generic models by combining data of PhCs and PCPs, PhCs and SHs, PCPs and SHs, and PhCs, PCPs, and SHs showed acceptable performance. The best performing combined model for the prediction of PhCs, PCPs, and SHs was based on area, HLR, OLR, and HRT (RMSEs: training set: 13%; test set: 22%). The information obtained by the use of these models may guide researchers, practitioners, policy makers, and citizens in enhancing knowledge and understanding for the design and operation of CWs in the field conditions. The objective of this study was to estimate the risk of potential adverse drug events (ADEs) associated with drugs repurposed for coronavirus disease 2019 (COVID-19) treatment in a large-scale population. Drug claims were used to calculate a baseline medication risk score (MRS) indicative of ADE risk level. Fictitious claims of repurposed drugs were added, one at a time, to patients drug regimens to calculate a new MRS and compute a level of risk. Drug claims data from enrollees with Regence health insurance were used and sub-payer analyses were performed with Medicare and com. insured groups. Simulated interventions were conducted with hydroxychloroquine and chloroquine, alone or combined with azithromycin, and lopinavir/ritonavir, along with terfenadine and fexofenadine as pos. and neg. controls for drug-induced Long QT Syndrome (LQTS). There were 527,471 subjects (56.6women; mean [SD] age, 47 years [21]) were studied. The simulated addition of each repurposed drug caused an increased risk of ADEs (median MRS increased by two-to-seven points, p < 0.001). The increase in ADE risk was mainly driven by an increase in CYP450 drug interaction risk score and by drug-induced LQTS risk score. The Medicare group presented a greater risk overall compared to the com. group. All repurposed drugs were associated with an increased risk of ADEs. Our simulation strategy could be used as a blueprint to preemptively assess safety associated with future repurposed or new drugs. According to current guidelines, oral antihistamines are the first-line treatment for chronic spontaneous urticaria (CSU). Up-dosing antihistamines to 4-fold the licensed dose is recommended if control is not achieved. Such indications are based mainly on expert opinion. To critically review and analyze clin. evidence on the efficacy and safety of higher-than-licensed dosage of second-generation oral antihistamines in the treatment of CSU. A systematic literature review was performed following a sensitive search strategy. All articles published in PubMed, EMBASE, and the Cochrane Library between 1961 and Oct. 2018 were examined Publications with CSU patients prescribed secondgeneration antihistamines in monotherapy compared with placebo, licensed dosages, and/or higher dosages were included. Articles were evaluated by peer reviewers. Quality was evaluated using the Jadad and Oxford scores. We identified 337 articles, of which 14 were included in the final evaluation (fexofenadine, 6; cetirizine, 2; levocetirizine and desloratadine, 1; levocetirizine, 1; rupatadine, 2; ebastine, 1; and bilastine, 1). Only 5 studies were placebo-controlled. The number of patients included ranged from 20 to 439. The observation lapse was ≤16 wk. High fexofenadine doses produced a significant dosedependent response and controlled urticaria in most patients. Cetirizine, levocetirizine, rupatadine, and bilastine were more effective in up-dosing. The most frequent adverse events were headache and drowsiness.
