The reaction is accomplished under mild conditions without the use of a corrosive acid or base as an additive. The Cl exchange reaction of hexachlorocyclotriphosphazene, N3P3Cl6 (1), with one equimolar amount of sodium salt of N/O donor type bidentate ligand containing a 2-pyridyl pendant arm (2) afforded, regioselectively, the partly substituted 2-pyridyl(N/O)spirocyclotriphosphazene (3; with a yield of 65%) in THF. The reactions of 3 with excess pyrrolidine, morpholine and 1,4-dioxa-8-azaspiro[4,5]decane (DASD) led to the formation of the tetraamino-2-pyridyl(N/O)spirocyclotriphosphazenes (3a-3c) in high yields. Compound 3 also gave both tetrapiperidino (3d) and gem-bispiperidino (3e) products with excess piperidine. The structures of all the compounds were determined by elemental analyses, ESI-MS, FTIR, HSQC, HMBC and H-1, C-13, and P-31 NMR techniques. The crystal structure of 3c was identified by single crystal X-ray crystallography. Besides, the compound 3e had one stereogenic P atom, and its chirality was verified by P-31 NMR spectroscopy in the presence of (S)-(+)-2,2,2-trifluoro-1-(9′-anthryl)-ethanol (CSA).

This report describes a carboxylate-assisted palladium-catalysed Catellani reaction, which is compatible with ortho-amination and unactivated C(sp(2))-H arylation. This method was used to synthesize a series of 1-amino substituted dihydrophenanthridines, phenanthridines and 6H-benzo[c]chromenes. Based on kinetic isotope experiments, the kinetic curve proves that pivalic acid accelerates the reaction rate of unactivated C(sp(2))-H activation, and thus this rate can keep up with the five membered aryl-norbornene-palladacycle (ANP) intermediate.

We report on the discovery of norbornyl moiety as a novel structural motif for cyclin-dependent kinase 2 (CDK2) inhibitors which was identified by screening a carbocyclic nucleoside analogue library. Three micromolar hits were expanded by the use of medicinal chemistry methods into a series of 16 novel compounds. They had prevailingly micromolar activities against CDK2 and the best compound of the series attained IC(50)of 190 nM. The binding modes were explored in molecular details by modeling and docking. Quantum mechanics-based scoring was used to rationalize the affinities. In conclusion, the discovered 9-hydroxymethylnorbornyl moiety was shown by joint experimental-theoretical efforts to be able to serve as a novel substituent for CDK2 inhibitors. This finding opens door to the exploration of chemical space towards more effective derivatives targeting this important class of protein kinases.

Highly functionalized 4-aminoindoles were synthesized via the three-component cross-coupling of o-iodoaniline, N-benzoyloxyamines, and norbornadiene. The Catellani and retro-Diels-Alder strategy was used in this domino process. o-Iodoaniline, with electron-donating and sterically hindered protecting groups, made the reaction selective toward o-C-H amination. On the basis of density functional theory calculations, the intramolecular Buchwald coupling of this reaction underwent a dearomatization and a 1,3-palladium migration process. The reasons for the control of the chemical selectivity by the protecting groups are given. Moreover, synthetic applications toward 4-piperazinylindole and a GOT1 inhibitor were realized.
